male
sterility
and
histoincompatibility
mutant
,
mshi
,
results
from a
recessive
mutation
that
arose
spontaneously
in the
BALB/cBy
inbred
mouse
strain
.
Using
a
402-member
BALB/cBy-mshi/mshi
(BALB/cBy-mshi
C57BL/6J)
F1
backcross
panel
, the
mutation
has been
mapped
on
proximal
mouse
chromosome
10
,
between
markers
D10Mit51/168/212
and
D10Mit213
. There are
nineteen
genes
between
these
two
markers
. The
IL20RA
gene
is
in this
critical
interval
, and
it
encodes
a
cell
surface
receptor
that
is
highly
expressed
in
skin
and
testes
. These
characteristics
make
IL20RA
a
good
candidate
for the
gene
that
is
altered
in the
mshi
mutation
. To
determine
if
IL20RA
is
the
molecular
basis
of
mshi
,
we
sequenced
the
IL20RA
gene
within
two
different
wild-type
,
heterozygous
, and
homozygous
mutant
mice
. There
is
no
base
pair
change
between
wild-type
BALB/cBy
and
mutant
mice
.
One
polymorphism
was
found
between
the
two
different
wild-type
mice
in
coding
regions
and
several
changes
in
non-coding
regions
were also
noted
.
Using
one
of these
single
nucleotide
polymorphisms
between
BALB/cBy
and
C57BL/6J
, the
402
member
backcross
panel
was
typed
for
BALB/cBy-derived
versus
C57BL/6J-derived
alleles
of
IL20RA
.
Five
crossovers
separated
IL20RA
and
mshi
,
placing
IL20RA
about
1.2
cM
centromeric
of
mshi
. The
region
between
IL20RA
and
D10MIR213
–
where
mshi
is
now
restricted
–
includes
only
7
genes
.
