that are
hypotrichotic
(lacking
normal
hair)
due
to the
autosomal
recessive
shorn
(shn)
mutation
showed
a
low
survival
rate
in
brother
by
sister
matings
in a
breeding
colony
of
SU/Csu
rats
. The
majority
of
rats
died
before
weaning
.
Subsequent
breedings
of
female
SU/Csu
x
Brown
Norway
F1
hybrids
with
normal
hair
(shn/+)
to
shn/shn
SU/Csu
male
rats
, and the
reciprocal
cross
,
indicated
that the
lack
of
survival
was
due
to the
mothers
shnr/shn
genotype
rather
than the
pup's
genotype
. This
observation
suggested
that, in
addition
to
hairlessness
,
shd/shn
may
be
responsible
for "
maternal
insufficiency
" in
homozygous
females
. A "
forced
heterozygous
"
breeding
plan
was
established
with the
line
of
rats
exhibiting
the
shorn
mutation
,
where
heterozygous
females
(shn/+)
with
hair
were
crossed
with
homozygous
(shd/shn)
males
to
maintain
the
shorn
line
. For
four
generations
, this
produced
a
significantly
larger
litter
size
, as
anticipated
. By the
fifth
generation
,
however
,
heterozygous
mothers
also
showed
maternal
insufficiency
resulting
in
reduced
litters
. This
genetic
separation
of the
maternal
insufficiency
from the
hypotrichotic
phenotype
indicated
that these
may
be
separate
traits
, and not a
pleotropic
effect
of the
same
mutation
.
Concurrently
,
similar
backcrosses
of
F1
hybrids
of
Brown
Norway
rats
and
Charles
River
hairless
rats
(another
hypotrichotic
line
of
rats)
were
set
up
to
try
to
determine
if
maternal
insufficiency
was
shorn-specific
or
else
was
simply
a
result
of
having
no
hair
.
Reciprocal
crosses
resulted
in
similar
reduced
litters
where
the
mother
had
no
hair
. In
summary
,
my
results
seem
to
indicate
that
maternal
insufficiency
in the
shorn
rats
is
a
separate
trait
from
hypotrichosis
. This
suggests
that
it
should be
possible
,
even
if these
two
traits
are
linked
, to
select
for a
line
with
shn-generated
hypotrichosis
, but not the
maternal
insufficiency
trait
. By
April
10,2005
,
backcrossing
to
wild-type
RR
rats
produced
one
shn/shn
mother
which
was
tested
and she
reduced
from
around
twelve
down
to
zero
pups
before
weaning
. The
results
of the
Charles
River
crosses
appear
to
indicate
the
possibility
of
some
involvement
of
maternal
hypotrichosis
in the
litter
reduction
seen
in this
line
. The
litter
reduction
seen
in
Charles
River
hairless
may
, or
may
not have a
cause
distinct
from that
seen
in
Shorn
rats
.
