Title
Purification
and
characterization
of an
antimicrobial
compound
secreted
by a
soil
bacterium
Publisher
Central Connecticut State University
Date
2008
Resource Type
Master's Thesis
Notes
The
discovery
of
antibiotics
was
one
of the
greatest
discoveries
of the
21st
20th
century
(Van
Kraaig
,
1999
and
Livermore
,
2004)
. The
current
need
for
new
antibiotics
to
fight
bacterial
pathogens
that have
become
resistant
is
growing
every
day
. In this
work
we
characterize
and
partially
purify
an
antibacterial
compound
that
is
produced
by a
soil
bacterium
.
Bacterial
strain
"
Unknown
Dirt
" was
isolated
in the
Davis
laboratory
from a
soil
sample
collected
in
central
Connecticut
. This
strain
was a
particularly
interesting
candidate
because
it
was
able
to
inhibit
the
growth
, in
co-cultivation
experiments
, of
Mycobacterium
smegmatis
, a
relative
of
Mycobacterium
tuberculosis
. The
strain
has also been
shown
to
inhibit
other
bacteria
such
as
B
.
megaterium
and
E
.
coli
. In this
work
we
characterize
the
secreted
molecule
("UD")
as a
high
molecular
weight
compound
through
dialysis
experiments
. The
data
also
suggest
that
UD
is
protein
in
nature
.
Finally
we
determine
the
genus
and
species
that
produces
the
antibacterial
compound
UD
.
