Abstract
RNA
editing
is
a
mechanism
which
is
used
to
increase
the
number
of
possible
protein
products
from a
specific
gene
. The
catalytic
enzymes
for
RNA
editing
are
known
to be
present
in a
wide
variety
of
organisms
,
including
many
different
insects
.
It
has been
shown
that the
frequency
of
RNA
editing
fluctuates
between
the
following
stages
of
development
,
egg/larva
,
larva
,
pupae
, and
adult
.
Our
goal
was to
use
a
known
editing
site
and
measure
the
frequency
of
editing
as the
adult
fly
ages
.
A-to-I
RNA
editing
is
an
enzymatic
process
in
which
Adenosine
is
converted
to
Inosine
post-transcriptionally
by the
ADAR
family
of
enzymes
.
Analysis
has been
done
in the
Slowpoke
potassium
ion
channel
to
observe
the
frequency
at
which
RNA
editing
of this
transcript
occurs
. In
order
to
detect
RNA
editing
using
sequencing
,
it
is
necessary
to
convert
RNA
transcripts
to
DNA
products
for
sequencing
.
RT-PCR
is
a
technique
that
utilizes
the
enzyme
reverse
transcriptase
to
convert
an
RNA
molecule
into a
complementary
DNA
(cDNA)
molecule
that
can
then be
used
as a
template
for a
PCR
reaction
. The
PCR
product
may
then be
sequenced
.
RNA
editing
is
detectable
as
mixed
signals
in
sequencing
chromatograms
, with
controls
to
distinguish
editing
from
DNA
polymorphism
.
Through
this
research
it
was
determined
that the
frequency
at
which
the
RNA
is
edited
in the
Slowpoke
potassium
ion
channel
shows
an
increase
from
young
adult
flies
(1-3
days
after
hatching)
to
old
adults
(35-36
days
after
hatching)
.
Ultimately
the
goal
is
to
obtain
RNA
editing
sites
unique
to
single
insect
species
that
could
then be
used
for the
possible
development
of
species-specific
pesticides
.
